TRex Bio, Inc. develops biologic drugs that regulate the immune system, for autoimmune and inflammatory diseases, in the United States and abroad. Its Deep Biology Platform is used to discover and design drugs by combining tissue-level knowledge of human biology and Treg function to find and study targets. It relies on computational mapping, which uses in silico tools, a human tissue bank and an atlas of Treg gene expression to trace immune regulation pathways in tissue.

The platform also includes the Treg Atlas, which recreates tissue states in Tregs and reprograms Tregs taken from blood to mimic tissue-specific pathways in the lab for target validation. A third part, the Phenotypic Assay Cascade, is a set of high-throughput functional assays that test targets across donors and inflammatory settings.

Its lead candidate, TRB-061, is an investigational TNFR2 agonist built as an Fc-fusion protein, meant to activate and expand Tregs in inflamed tissue. TRB-071 is an agonist for inflammatory bowel disease designed to boost tissue Tregs and block an inflammatory signaling pathway. TRB-051, a partnered modulator of immune effector cells, targets autoimmune and inflammatory diseases including lupus affecting the skin.

The company also has preclinical programs on tissue Treg biology guided by its platform. It was incorporated in 2018 and is based in South San Francisco, California.
